Starting from Ved 1.8.0, it's now possible to playtest from Ved easily! To test levels in VVVVVV (and not in VVVVVV-CE), this does require having an updated version of VVVVVV (2.3) which isn't yet officially released. It's possible to compile a state-of-the-art version of VVVVVV yourself, but if you don't want to do that, here's some recent executables for the Make and Play version.

This is an intermediate version of VVVVVV Make and Play 2.3, built based on the files from the VVVVVV repository.

Why not use VVVVVV-CE for testing VVVVVV levels? First of all, VVVVVV-CE stopped being worked on, and the next sentences were written before that. It's better to test VVVVVV levels on VVVVVV to make sure you haven't accidentally done something that VVVVVV-CE accepts but VVVVVV doesn't. VVVVVV-CE was made to support levels made for VVVVVV, but not to emulate that its changes (like removed limits) weren't made when a level is played that doesn't target VVVVVV-CE. For example, you could accidentally go over the script lines limit or use internal scripting commands in a simplified script and inadvertently release a VVVVVV-CE-only level!

For reference, VVVVVV Make and Play 2.2 can be downloaded from here: https://thelettervsixtim.es/makeandplay/. You probably want to store the intermediate version of 2.3 in its own folder. On Windows, you only need to add data.zip, on other operating systems, download a fresh copy of 2.2 and replace it as described:

Windows: Just add data.zip. If you don't have one at hand, you can simply get it from VVVVVV M&P 2.2.
macOS: This one may actually require a bit more work, sorry. But if you copy-paste Frameworks, Resources and Info.plist from the package contents of an existing VVVVVV .app, it should work. Alternatively, copy MacOS/VVVVVV and paste it in place of VVVVVV in an existing VVVVVV .app.
Linux: Assuming Ubuntu 18.04; another version or distro might work. Last I checked, the downloadable version of VVVVVV is an installer, so you probably want to copy the ~/vvvvvv folder. Only the VVVVVV binary is supplied, this can be used as a drop-in replacement of your existing binary. Probably ~/vvvvvv/x86_64/vvvvvv.x86_64.
